how to remove "dev~" before sitenane in URL?
e.g. sitename is "cms", i want url to be
http://example.com/cms/
instead of
http://example.com/dev~cms/

It can not, because then we will not know whether "CMS" is just a page name or a site name or others.

However you do can configure www.kooboosite.com into the CMS website and use www.kooboosite.com to visit and develop it.

scenerio:
server: iis6 with .net 4.0 installed.
application path: http://hostname/web/myapp/, where myapp is a legacy asp.net (mvc1.0) web application,
i want to keep the "myapp" unchanged, and ftp upload kooboo to this folder, replace the old app,
the OLD virtual folder URL should not be changed,
is this possible?

now i want to be:
http://sitename.com
instead of
http://sitename.com/dev~mysite

is possible?

i mean i want to set Site mysite to Default site when ppl go to http://sitename.com
